Chihayafuru adalah anime drama sekolah bertemakan permainan tradisional Jepang yang disebut Karuta. Karuta merupakan permainan kartu yang terinspirasi dari antologi klasik Jepang "100 Puisi". Para pemain berhadapan satu sama lain untuk saling merebut kartu berisi puisi yang disusun di antara mereka. Dibutuhkan kecepatan tangan dan ketajaman pendengaran demi memenangkan game ini.

Ayase Chihaya adalah gadis tomboy yang selalu bersemangat dan punya kemauan keras. Sayangnya, sifat cerianya tak bisa menutupi kelemahan bahwa dirinya tidak mempunyai impian atau prestasi yang bisa dibanggakan, membuatnya merasa rendah dibandingkan kakaknya yang seorang model. Di sekolah ia berteman baik dengan Mashima Taichi, yang sebenarnya menaruh hati pada Chihaya.

Tapi perasaan itu tak pernah berbalas terlebih setelah Chihaya berteman dengan Wataya Arata, seorang pemain karuta berbakat. Arata adalah cucu dari grandmaster Karuta, dan bermimpi mengejar tempat kakeknya itu dengan merebut gelar King, sebutan pemain Karuta terbaik untuk pria. Sejak saat itu, Chihaya jatuh cinta dengan dunia Karuta dan kehidupannya tak pernah lepas dari game itu.
